DiDomenico won re-election to the Middlesex, Suffolk and Essex District seat in 2010. Timothy Flaherty ran against him in the primary. The primary election took place on September 14, 2010. Barbara Bush ran for the seat on the Republican ticket. The general election took place on November 2, 2010.

2010 special election
On May 11, 2010, DiDomenico won a special election to represent the Middlesex, Suffolk and Essex Senate District in the Massachusetts State Senate, defeating independent candidate John Cesan.[8] The election was held to fill the seat made vacant by the resignation of Senator Anthony D. Galluccio.
