Chronic opioid receptor (OR) activation by morphine causes distinct cellular adaptations responsible for the development of tolerance. The present study examines the effect of chronic morphine exposure on the ability of high-efficacy agonists to mediate �-OR (DOR) and �-OR (MOR) uncoupling and internalization, two regulatory mechanisms contributing to rapid desensitization of OR function. To determine celiac disease (CD) prevalence and associated manifestations or risk factors in healthy adult Emiratis. We identified nine naturally-occurring human single nucleotide polymorphisms (SNPs) in the alpha(1a)-adrenoceptor (alpha(1a)AR) coding region, seven of which result in amino acid change. Utilizing rat-1 fibroblasts stably expressing wild type alpha(1a)AR or each SNP at both high and low levels, we investigated the effect of these SNPs on receptor function. Compared with wild type, two SNPs (R166K, V311I) cause a decrease in binding affinity for agonists norepinephrine, epinephrine, and phenylephrine, and also shift the dose-response curve for norepinephrine stimulation of inositol phosphate (IP) production to the right (reduced potency) without altering maximal IP activity. Progression through mitosis occurs because cyclin B/Cdc2 activation induces the anaphase promoting complex (APC) to cause cyclin B destruction and mitotic exit. To ensure that cyclin B/Cdc2 does not prematurely activate the APC in early mitosis, there must be a mechanism delaying APC activation. Emi1 is a protein capable of inhibiting the APC in S and G2. We show here that Emi1 is phosphorylated by Cdc2, and on a DSGxxS consensus site, is subsequently recognized by the SCF(betaTrCP/Slimb) ubiquitin ligase and destroyed, thus providing a delay for APC activation. Failure of betaTrCP-dependent Emi1 destruction stabilizes APC substrates and results in mitotic catastrophe including centrosome overduplication, potentially explaining mitotic deficiencies in Drosophila Slimb/betaTrCP mutants. We hypothesize that Emi1 destruction relieves a late prophase checkpoint for APC activation.

Although the Rap1A protein resembles the oncogenic Ras proteins both structurally and biochemically, Rap1A exhibits no oncogenic properties. Rather, overexpression of Rap1A can reverse Ras-induced transformation of NIH 3T3 cells. Because the greatest divergence in amino acid sequence between Ras and Rap1A occurs at the COOH terminus, the role of this domain in the opposing biological activities of these proteins was examined. Lipid droplets (LDs) are the major fat storage organelles in eukaryotic cells, but how their size is regulated is unknown. Using genetic screens in C. elegans for LD morphology defects in intestinal cells, we found that mutations in atlastin, a GTPase required for homotypic fusion of endoplasmic reticulum (ER) membranes, cause not only ER morphology defects, but also a reduction in LD size. Similar results were obtained after depletion of atlastin or expression of a dominant-negative mutant, whereas overexpression of atlastin had the opposite effect. Atlastin depletion in Drosophila fat bodies also reduced LD size and decreased triglycerides in whole animals, sensitizing them to starvation. In mammalian cells, co-overexpression of atlastin-1 and REEP1, a paralog of the ER tubule-shaping protein DP1/REEP5, generates large LDs. Purpose: The study aimed to evaluate the (i) regulatory role of G i-like protein during the acrosome reaction (AR) of normal sperm donors and (ii) the role of intact acrosomes during sperm–zona binding. Methods: The acrosomal exocytosis of spermatozoa incubated with solubilized zona pellucida (ZP) at a final concentration of 1 ZP/μl was compared with 10 μM calcium ionophore A23187 and 30% (v/v) pooled human follicular fluid (HFF). Spermatozoa were incubated with 1, 10, and 100 ng/ml pertussis toxin (PT) during capacitation to functionally inactivate the G i-like protein. The sperm–zona binding potential of 100 ng/ml PT-treated spermatozoa followed, by exposure to 1 ZP/μl, revealed significantly higher zona-bound spermatozoa compared to controls treated with 1 ZP/μl only. Results: PT treatment of spermatozoa did not affect sperm motility, however, inhibited the percentage AR induced by solubilized ZP. In contrast, the A23187- and HFF-induced ARs were not sensitive to PT treatment. PT inhibition of the ZP-induced AR occurred in a concentration-dependent manner, with maximal effects observed at 100 ng/ml PT. Conclusions: In conclusion, it seems that PT-sensitive G i-like protein in human spermatozoa plays an important regulatory role in the AR induced by the human ZP, and this underlines the importance of intact acrosomes during sperm–zona binding.
