Phenotypes

Recent enhancement of the pool of known molecular markers for avocado has allowed the construction of the first moderately dense genetic map for this species. Over 300 SSR markers have been characterized and 163 of these were used to construct a map from the reciprocal cross of two Florida cultivars 'Simmonds' and 'Tonnage'. One hundred thirty-five primer pairs amplified 163 usable loci with 20 primer pairs amplifying more than one locus. 'Tonnage' was heterozygous for 152 (93%) loci, whereas 'Simmonds' was heterozygous for 64 (39%). Null alleles were identified at several loci. Linkage maps were produced for both reciprocal crosses and combined to generate a composite linkage map for the F1 population of 715 individuals. The composite map contains 12 linkage groups. Linkage groups ranged in size from 157.3 cM (LG2) to 2.4 cM (LG12) and the number of loci mapped per group ranged from 29 (LG1) to two (LG12). The total map length was 1,087.4 cM. Only seven markers were observed to have segregation distortion (α ≤ 0.05) across both sub-composite (reciprocal) maps. Phenotypic data from traits of horticultural interest are currently being collected on this population with the ultimate goal of identifying useful quantitative trait loci and the development of a marker-assisted selection program.

The aim of this study was to investigate the adhesive phenotype of the human intestinal isolate Bifidobacterium bifidum MIMBb75 to human colon carcinoma cell lines. We have previously shown that the adhesion of this strain to Caco-2 cells is mediated by an abundant surface lipoprotein named BopA. In this study, we found that this strain adheres to Caco-2 and HT-29 cells, and that its adhesion strongly depends on the environmental conditions, including the presence of sugars and bile salts and the pH. Considerably more adhesion to a Caco-2 monolayer occurred in the presence of fucose and mannose and less when MIMBb75 grew in Oxgall bile salts compared to standard environmental conditions. In particular, growth in Oxgall bile salts reduced the adhesion ability of MIMBb75 and modified the SDS-PAGE profile of the cell wall associated proteins of the strain. The pH markedly affected both adhesion to Caco-2 and bacterial autoaggregation. Finally, experiments with sodium metaperiodate suggested that not only proteinaceous determinants are involved in the adhesion process of B. bifidum. In conclusion, it seems that the colonization strategy of this bacterium can be influenced by factors varying along the gastrointestinal tract, such as the presence of specific sugars and bile salts and the pH, possibly limiting the adhesion of B. bifidum to only restricted distal sites of the gut.

Fascioliasis, caused by liver fluke species of the genus Fasciola, has always been well recognized because of its high veterinary impact but it has been among the most neglected diseases for decades with regard to human infection. However, the increasing importance of human fascioliasis worldwide has re-launched interest in fascioliasis. From the 1990s, many new concepts have been developed regarding human fascioliasis and these have furnished a new baseline for the human disease that is very different to a simple extrapolation from fascioliasis in livestock. Studies have shown that human fascioliasis presents marked heterogeneity, including different epidemiological situations and transmission patterns in different endemic areas. Presently, wildcatFelis silvestris silvestris Schreber, 1777 populations are fragmented and rapidly declining in most of Europe. Although habitat destruction possibly constitutes the most serious threat to wildcat survival, hybridisation with feral domestic cats is also a critical problem. However, the mechanisms that allow domestic cats to colonise former wild cat home ranges are yet unclear. The present paper describes the decrease of typical phenotypic wildcats and the increase of phenotypic domestic cats in a remote wild area of Portugal (Serra da Malcata). A field survey using box-traps and radio-tracking between 1998 and 2001 revealed that wildcats were widespread in the study area and no domestic cats were present. A second survey using camera traps between 2005 and 2007 revealed only one wildcat whereas four typical domestic phenotype individuals were photographed. The present study clearly emphasizes the need for urgent measures aimed at preserving wildcat populations. These measures should include a national census of the species and an extensive monitoring of genetic integrity of wildcat populations, followed by the elaboration of a wildcat conservation action plan.

Aims:  Grey mould caused by Botrytis cinerea is an economically important disease of strawberries in Tunisia and worldwide. The aim of this study was to select effective halophilic bacteria from hypersaline ecosystems and evaluate the abilities of antifungal bacteria to secrete extracellular hydrolytic enzymes, anti-Botrytis metabolites and volatiles.Methods and Results:  Grey mould was reduced in strawberry fruits treated with halophilic antagonists and artificially inoculated with B. cinerea. Thirty strains (20·2%) were active against the pathogen and reduced the percentage of fruits infected after 3 days of storage at 20°C, from 50% to 91·66%. The antagonists were characterized by phenotypic tests and 16S rDNA sequencing. They were identified as belonging to one of the species: Virgibacillus marismortui, B. subtilis, B. pumilus, B. licheniformis, Terribacillus halophilus, Halomonas elongata, Planococcus rifietoensis, Staphylococcus equorum and Staphylococcus sp. The effective isolates were tested for antifungal secondary metabolites.Conclusions:  Moderately halophilic bacteria may be useful in biological control against this pathogen during postharvest storage of strawberries.Significance and Impact of the study:  The use of such bacteria may constitute an important alternative to synthetic fungicides. These moderate halophiles can be exploited in commercial production and application of the effective strains under storage and greenhouse conditions.

Ophrys orchids mimic the female sex pheromones of their pollinator species to attract males for pollination. Reproductive isolation in Ophrys is based on the selective attraction of only a single pollinator species. A change of floral odour can result in the attraction of a new pollinator species that acts as an isolation barrier towards other sympatrically occurring Ophrys species. Ophrys lupercalis, Ophrys bilunulata, and Ophrys fabrella grow sympatrically and bloom consecutively on Majorca and are pollinated by three species of Andrena. We investigated variation of phenotypic and genotypic flower traits, aiming to study the role of the floral odour for reproductive isolation and speciation. Using chemical and electrophysiology (gas chromatography coupled with an electroantennographic detector) methods, we show that the three Ophrys species use the same odour compounds for pollinator attraction, but in different proportions. A comparison of the floral odour bouquets in a multivariate analysis revealed a clear grouping of plants from the same species, although with an overlap between species. A comparison of the same plants using molecular markers gave a contrasting result. Although O. lupercalis and O. fabrella were genetically well separated, plants of O. bilunulata did not form a distinct group but were similar to either O. lupercalis or O. fabrella. Our data indicate gene flow and hybridization to occur between O. bilunulata and O. lupercalis as well as between O. bilunulata and O. fabrella. All plants of O. bilunulata, despite having different genotypes, showed a very similar floral odour. This reflects a strong selective pressure by the pollinating males. The overlap of genotypes of O. bilunulata and O. fabrella supports our hypothesis that O. fabrella diverged from O. bilunulata by scent variation and the attraction of a new pollinator species, Andrena fabrella. © 2009 The Linnean Society of London, Biological Journal of the Linnean Society, 2009, 98, 439–451.

The great majority of phenotypic characteristics are complex traits, complicating the identification of the genes underlying their expression. However, both methodological and theoretical progress in genome-wide association studies have resulted in a much better understanding of the underlying genetics of many phenotypic traits, including externally visible characteristics (EVCs) such as eye and hair color. Consequently, it has become possible to predict EVCs from human samples lacking phenotypic information. Predicting EVCs from genetic evidence is clearly appealing for forensic applications involving the personal identification of human remains. Now, a recent paper has reported the genetic determination of eye and hair color in samples up to 800 years old. The ability to predict EVCs from ancient human remains opens up promising perspectives for ancient DNA research, as this could allow studies to directly address archaeological and evolutionary questions related to the temporal and geographical origins of the genetic variants underlying phenotypes.The great majority of phenotypic characteristics are complex traits, complicating the identification of the genes underlying their expression. A method for the separation of the beta-LG variants A, B, and C was developed using free zone capillary electrophoresis. A separation buffer consisting of 50 mM 2-(N-morpholino)ethane sulfonic acid (pH 8.0) and polyoxyethylene (20)-sorbitan monolaurate was used for the separation. Milk samples from 424 Jersey cows were phenotyped using this method, and the gene frequencies for beta-LG A, B, and C were .41, .53, and .06, respectively. These frequencies were different from those found in studies of Jersey populations in other countries, where the beta-LG B allele had a significantly higher frequency, and the beta-LG A allele a significantly lower frequency, than those of this present study. The frequency of the beta-LG C allele was intermediate to those of other studies. The concentration of beta-LG in the milk was different among the beta-LG phenotypes and was, from greatest to least: AA, AB, AC, BB, and BC.

The article presents data from guidelines, consensus and literature sources on the modern point of view regarding to the pathogenesis, diagnosis and principles of treatment of women with polycystic ovary syndrome. It is a systemic pathology that occurs in women of any age, from puberty to menopause, involving almost all systems of the body in the process.
The work focuses on the variability of the clinical manifestations of this syndrome, which is characterized by menstrual irregularities, infertility, polycystic changes in the ovaries according to ultrasound, dermatopathies and metabolic disorders. The main purpose of diagnosing the syndrome is to determine the severity of clinical manifestations, the sources and pathogenesis of androgen hyperproduction, the impact on reproductive function, as well as the assessment of metabolic and cardiovascular risks.
Given the multifaceted clinical manifestations, the management of women with polycystic ovary syndrome requires a multidisciplinary approach, and pathogenetic therapy should include normalization of the hormonal profile and menstrual function, treatment of dermatopathies, correction of metabolic disorders, treatment of infertility (if pregnancy is relevant), etc.
