Triclad flatworms are well studied for their regenerative properties, yet little is known about their embryonic development. We here describe the embryonic development of the triclad Schmidtea polychroa, using histological and immunocytochemical analysis of whole-mount preparations and sections. During early cleavage (stage 1), yolk cells fuse and enclose the zygote into a syncytium. The zygote divides into blastomeres that dissociate and migrate into the syncytium. During stage 2, a subset of blastomeres differentiate into a transient embryonic epidermis that surrounds the yolk syncytium, and an embryonic pharynx. Other blastomeres divide as a scattered population of cells in the syncytium. During stage 3, the embryonic pharynx imbibes external yolk cells and a gastric cavity is formed in the center of the syncytium. The syncytial yolk and the blastomeres contained within it are compressed into a thin peripheral rind. From a location close to the embryonic pharynx, which defines the posterior pole, bilaterally symmetric ventral nerve cord pioneers extend forward. Stage 4 is characterized by massive proliferation of embryonic cells. Large yolk-filled cells lining the syncytium form the gastrodermis. During stage 5 the external syncytial yolk mantle is resorbed and the embryonic cells contained within differentiate into an irregular scaffold of muscle and nerve cells. Epidermal cells differentiate and replace the transient embryonic epidermis. Through stages 6–8, the embryo adopts its worm-like shape, and loosely scattered populations of differentiating cells consolidate into structurally defined organs. Our analysis reveals a picture of S. polychroa embryogenesis that resembles the morphogenetic events underlying regeneration.

The purpose of this work was to evaluate the in vitro virulence of three isolates of Metarhizium anisopliae var. anisopliae to eggs and larvae of the tick Boophilus microplus. The fungus tested was isolated from engorged females of B. microplus collected in the field, and identified as Ma01, Ma02 e Ma04. These isolates were evaluated by immersion of eggs and larvae in suspension with different conidial concentrations: 105, 106, 107 e 108 conidia/ml. In each isolate there was a treatment group for each spore concentration and a control group with 10 repetitions. It was observed in the treated egg groups that there was a hatching percentage that was much less than that observed in the control groups. This was in inverse proportion to the conidia concentration/ml. Larval bioassays of all the tested isolates resulted in a high mortality of larvae in direct proportion to the spore concentration/ml, 10 days after the conidia suspensions were inoculated. Aqueous leaf extract of Ricinus communis L (Euphorbiaceae), a cultivated plant in tropical countries, showed excellent insecticidal activity against Callosobruchus chinensis L (Coleoptera: Bruchidae). We have isolated and tested flavonoids as insecticidal and antimicrobial agents. The isolated flavonoids showed potential insecticidal, ovicidal and oviposition deterrent activities against C chinensis L. However, antimicrobial activity against the common microbial infestants of stored pulses, of which C chinensis is a major pest, was found to be insignificant. Two bands having Rf 0.63 and 0.69 were seen on HPTLC plates using mobile phase benzene + ethyl acetate + methanol + formic acid (12 + 4.5 + 2 + 1.5 by volume) as eluant. The Rf values and scanning of the spectrum in the ultraviolet region, showed identity with the flavonoids quercetin and kaempferol. This was further confirmed using HPLC and IR and UV spectrometry. HPLC and HPTLC chromatograms also suggested quercetin to be the major flavonoid present in the hydrolyzed aqueous leaf extract of R communis.

Two fractions showing proteolytic enzymes have been obtained from the latex of Synadenium grantii Hook, ‘f’, using gel-filtration and anion-exchange chromatographic techniques. Both these proteases have the same molecular mass of 76±2 kDa each. They exhibit maximal activity at pH 7.0 and at a temperature of 60 °C. They display stability over a pH range from 5–10 and are also highly thermostable. Irreversible inhibition by PMSF indicates that they are serine proteases. In addition, histidine residues also appear to play an important role in catalysis as evidenced by inhibition with DEPC. They also exhibit similarity with respect to pH and temperature optima, kinetic properties and thermal stability.

Presenting in her late twenties, this case report examines a G6P2 patient at 11-weeks gestation that was diagnosed with a blighted
ovum, as well as the subsequent outcome and methods of additional management. A blighted ovum refers to a fertilized egg that
does not develop, despite the formation of a gestational sac. The most common cause of a blighted ovum is of genetic origin.
Trisomies account for most first trimester miscarriages, while consanguineous marriages result in recurrent miscarriages due to
a blighted ovum. Additionally, a higher percentage of deoxyribonucleic acid (DNA) damage in sperm carries a higher rate of
miscarriage. Nutritional factors that may lead to a blighted ovum include low-levels of copper, prostaglandin E2, and anti-oxidative
enzymes. High body mass index (BMI), especially in women with a BMI≥30 kg/m2
has been shown to be linked to a blighted
ovum. Globally, it has been shown that a blighted ovum is a serious adverse event related to vaccination against dengue fever.

Background, aim and scope The biocide triclosan (TCS) is commonly used in personal care, acrylic, plastic, and textiles products. TCS has been detected in surface water in several countries, and its ecological impact is largely unknown. In this work, the toxicity of TCS in zebrafish (Danio rerio), embryos and adults was studied. Several lethal and sub-lethal endpoints were analysed in organisms exposed to TCS such as mortality, embryo development and behaviour, hatching, micronuclei and biochemical markers (cholinesterase (ChE), glutathione S-transferase (GST) and lactate dehydrogenase (LDH)). Materials and methods Embryo/larvae assay followed the OECD guideline on Fish Embryo Toxicity Test. Embryos were exposed at nominal concentrations of 0.1, 0.3, 0.5, 0.7 and 0.9 mg/l of TCS for 6 days and were inspected daily with the help of a stereomicroscopy for mortality, developmental parameters (otolith formation, eye and body pigmentation, somite formation, heart beat, tail circulation, detachment of the tail-bud from the yolk sac) and hatching. A similar test was run to obtain larvae for ChE, GST and LDH analysis. The adult test followed the OECD Guideline TG 203 in semi-static conditions. Adult zebrafish of similar length and age were exposed to nominal concentrations of 0.1, 0.2, 0.3, 0.4 and 0.5 mg/l of TCS for 96 h and were inspected daily for mortality and behaviour alterations. A second test was run to obtain organs for biomarkers analysis: Heads, muscles and gills were isolated and snap-frozen in eppendorfs and used for ChE, LDH and GST determinations, respectively. Adult zebrafish testing also comprised a third test for micronucleus analysis in which the nominal concentrations of 0, 0.175 and 0.350 mg/l were used. Peripheral blood was obtained by cardiac puncture and used for the analysis. Results TCS showed acute toxicity for embryo/larvae (96 h LC50 = 0.42 mg/l) and delayed hatching. Moreover, embryo toxicity was evident: Delay on the otolith formation and eye and body pigmentation were found, and malformations were also evident, including spine malformations, pericardial oedema and undersize. Biomarkers levels were affected: ChE and LDH activity were increased in larvae exposed to 0.25 mg/l, and GST activity was increased in larvae exposed to 0.25 and 0.35 mg/l. TCS also demonstrated acute toxicity to adult zebrafish (96 h LC50 = 0.34 mg/l). However, TCS did not change biomarkers levels and did not elicit a micronucleus in adults. Discussions Despite the fact that similar 96 h LC50 values have been found for D. rerio embryos and adults (0.42 and 0.34 mg/l, respectively), the embryo assay was much more informative, showing important effects at several levels, including teratogenic response, hatching delay and alteration of biomarker levels. TCS does not seem to be genotoxic for adult fish or to interfere with biomarkers levels at the concentrations tested. Conclusions TCS has deleterious effects on zebrafish adults and during early stages, (including embryotoxicity, hatching delay and alterations of biomarkers levels). The range of endpoints used on the embryo test allows an integrated analysis that contributes to a better understanding of the toxicity and mode of action of TCS. Recommendations and perspectives Future works should focus on a deeper investigation of TCS modes of action on zebrafish early-life stages. As embryo testing was revealed to be so informative, a refinement of the test could be made, including other endpoints such as different biochemical markers as well as DNA microarrays to assess a gene expression level for the effect of exposure to TCS. In the perspective of risk assessment, these endpoints should be explored in order to assess their usefulness as early warning signs and links should be sought between these short-term tests and effects of long-term exposures as it is observed in more realistic scenarios.

The present investigation demonstrates establishment of United Nations Environment Programme (UNEP) priority Persistent Organic Pollutants (POPs) using high-resolution gas chromatography–high-resolution mass spectrometry. Particularly, POPs analytical methods were established using native and 13C-labeled internal standards of HCHs, HCB, cyclodienes, chlordanes, DDTs, mirex, dioxin-like PCBs, PCDDs, and PCDFs by isotope dilution technique. The relative response factor for 6-point calibration curve native standards (18 replicate analysis) were in the ranges of 0.93–1.43 with relative standard deviation ranges from 1.68 to 4.96%. Instrument detection limit and instrument quantification limit was established for various POPs at femtograms. Concentrations of UNEP-POPs were measured in liver (n = 10) and egg (n = 10) of great cormorants and their major diet, gizzard shad (n = 2), collected in and around Tokyo, Japan. DDTs (ranges in liver and egg, respectively) were predominant accumulants (9800–310,000 and 9600–73,000) followed by dioxin-like PCBs (4500–69,000 and 7900–150,000), chlordanes (2600–16,000 and 700–4,800), cyclodienes (650–4600 and <1–1000), HCB (680–2800 and 180–590), HCHs (230–1800 and 120–490), PCDD/DFs (3.2–27 and 1.7–5.7) on nanogram per gram lipid basis. Concentrations (ranges) of POPs in gizzard shad were in the following order: DDTs (3900–16,000), chlordanes (3400–14,000), cyclodienes (340–1300), HCB (110–480), and HCHs (140–360) on nanogram per gram lipid basis.

Avian egg shape is generally explained as an adaptation to life history, yet we currently lack a global synthesis of how egg-shape differences arise and evolve. Here, we apply morphometric, mechanistic, and macroevolutionary analyses to the egg shapes of 1400 bird species. We characterize egg-shape diversity in terms of two biologically relevant variables, asymmetry and ellipticity, allowing us to quantify the observed morphologies in a two-dimensional morphospace. We then propose a simple mechanical model that explains the observed egg-shape diversity based on geometric and material properties of the egg membrane. Finally, using phylogenetic models, we show that egg shape correlates with flight ability on broad taxonomic scales, suggesting that adaptations for flight may have been critical drivers of egg-shape variation in birds.

The specificity of early sperm-zona interactions in vitro was studied using gametes from the mouse, rabbit, pig, and human. Mouse epididymal spermatozoa, capacitated in vitro, bound to the zonae of intact mouse oocytes and zona fragments removed mechanically from oocytes. When viewed with the scanning electron microscope (SEM), spermatozoa were found adherent more frequently to the external (cumulus) surface of the zonae than the internal (oocyte) surface.
