Paper out from Arvinas on pre-clinical characterization of Vepdegestrant (ARV-471). Vepdegestrant was granted Fast Track status by the FDA earlier this year, and is currently being assessed in Phase 3 as a monotherapy and combination therapy with Palbociclib (CDK4/6 inhibitor, Pfizer) in the VERITAC-2 and -3 trials, respectively. The pre-clinical data looks highly promising, with tumour shrinkage in in vivo models when combined with Palbociclib. FLX475: An Azetidine-Containing, First-in-Class CCR4 Candidate with a 72 h Human Half-life | https://lnkd.in/eTJeU-VZ RAPT Therapeutics' oral CCR4 inhibitor FLX475 (100 mg QD, human T1/2 = 72 h) appears to shrink EBV+ tumors both as a monotherapy and in combination with a PD-1 inhibitor in Ph. I/II clinical trials (NCT03674567) while being generally well-tolerated. This case study reviews the molecule's interesting indirect mechanism on cancers, stimulating T cells to attack tumors, as well as FLX475’s notable discovery story involving significant PK optimization, scaffold hopping to an azetidine core, CYP TDI mitigation, and interesting synthetic steps. Yesterday, STAT published an insightful article on the #radiopharmaceutical space. It is indeed a "boom time" for those of us working to develop #radioligandtherapy (#RLT), and it’s great to hear there is interest in radiopharma as a potential "next wave of therapeutics for oncology." We agree! The field is not without challenge as the short-lived nature of the isotopes requires synchronized supply chains and isotope generation. Several CDMOs have stepped up their investment in the space to create distributed manufacturing networks that are more resilient than centralized approaches. This commitment was highlighted in a recent article in Endpoints News. At ARTBIO, we’re developing a pipeline of alpha radioligand therapeutics with Pb212 – our Alphadirect technology provides an accessible and reliable source of Pb212.
