Mastering HPLC: 32 Essential Tips for Pharma Professionals to Ensure Smooth Analysis 1. Ensure that you have a proper understanding of the HPLC system before starting. Familiarize yourself with the instrument manual and any specific protocols or methods that need to be followed. 2. Check the condition of the HPLC system, including the pump, injectors, column, and detector. Any damaged or faulty parts should be repaired or replaced before starting. 3. Clean the system thoroughly, including all components, by rinsing with solvent and running a series of blank runs. This helps remove any contaminants that may affect the analysis. 4. Prepare and condition the HPLC column according to the manufacturer's instructions. This typically includes flushing with an appropriate solvent and ensuring that the column is properly packed. 5. Prepare the mobile phase solvents, ensuring that they are of high purity and suitable for the analysis. Use appropriate filters to remove any impurities or particulates. 6. Prepare the sample for analysis, ensuring that it is properly dissolved or suspended in the appropriate solvent and filtered if necessary to remove any particulates. 7. Set up the HPLC system parameters, including flow rate, gradient program, and detector settings. This should be done in accordance with the specific analysis requirements. 8.
